What the landmark study shows
Mealey and Meurs published a study in 2008 in the Journal of the American Veterinary Medical Association on the breed distribution of the ABCB1-1Δ mutation, known as MDR1, across more than 5,300 genotyped dogs in North America (study on Google Scholar). The Australian Shepherd ranks high on the list: around half of the dogs tested carried at least one copy of the mutation, just behind the Collie, which remains the most affected breed. This mutation impairs a protein that normally keeps certain molecules from crossing the blood-brain barrier, exposing carrier dogs to severe neurological effects at doses that pose no risk to a non-carrier.
Keeping treatments safe day to day
- Get an MDR1 genetic test done via a cheek swab before any higher-risk treatment — a simple, one-time step that settles the question for good.
- Always mention the breed to your vet before prescribing high-dose ivermectin, loperamide or certain anaesthesia protocols, which are particularly relevant to this sensitivity.
- Never reuse an antiparasitic treatment prescribed for another dog, even from the same family, without first checking it's suitable for a carrier of the mutation.
